GLIA 9:176--187 (1993) Rat Ciliary Neurotrophic Factor (CNTF): Gene Structure and Regulation ofmRNA Levels in Glial Cell Cultures
نویسندگان
چکیده
The structure of the rat ciliary neurotrophic factor (CNTF) gene and the regulation ofCNTF mRNA levels in cultured glial cells were investigated. The rat mRNA is encoded by a simple two-exon transcription unit. Sequence analysis of the region upstream of the transcription start-site did not reveal a typical TATA-box consensus sequence. Low levels of CNTF mRNA were detected in cultured Schwann cells, and CNTF mRNA was not increased by a variety of treatments. Three-week-old astrocyteenriched cell cultures from new-born rat brain contained easily detectable CNTF mRNA. In astrocyte-enriched cultures, upregulation of CNTF mRNA levels was observed after treatment with IFN-"{. CNTF mRNA levels were down-regulated in these cells by treatments that elevate intracellular cyclic AMP and by members of the fibroblast growth factor (FGF) family. The implications of these results for potential in vivo functions of CNTF are discussed. © 1993 Wiley-Liss, Inc.

BMP and LIF signaling coordinately regulate lineage restriction of radial glia in the developing forebrain.
The earliest radial glia are neural stem cells that guide neural cell migration away from ventricular zones. Subsequently, radial glia become lineage restricted during development before they differentiate into more mature cell types in the CNS. We have previously shown that subpopulations of radial glial cells express markers for glial and neuronal restricted precursors (GRPs and NRPs) in expr...
متن کاملCiliary neurotrophic factor prevents retrograde neuronal death in the adult central nervous system.
The neurocytokine ciliary neurotrophic factor (CNTF) was described originally as an activity that supports the survival of neurons of the chicken ciliary ganglia in vitro. The widespread expression of CNTF and its principal binding protein, CNTF receptor alpha, in the central and peripheral nervous systems suggests a broader trophic role for this peptide. In the present study, we report that CN...
متن کاملAxotomy increases CNTF receptor mRNA in rat spinal cord.
In order to study the role of ciliary neurotrophic factor (CNTF) and its receptor (CNTF-R) in the response of spinal cord neurons to axotomy, we measured the levels of CNTF mRNA in nerve and CNTF-R mRNA in spinal cord following transection of sciatic nerve, using reverse transcriptase PCR. We found CNTF mRNA levels in the nerve fell and that CNTF-R mRNA levels in spinal cord increased at both 1...
متن کاملType-2 astrocyte development in rat brain cultures is initiated by a CNTF-like protein produced by type-1 astrocytes.
O-2A progenitor cells are bipotential glial precursors that give rise to both oligodendrocytes and type-2 astrocytes on a precise schedule in the rat CNS. Studies in culture suggest that oligodendrocyte differentiation occurs constitutively, while type-2 astrocyte differentiation requires an exogenous inducer such as fetal calf serum. Here we describe a rat brain cell culture system in which ty...
متن کاملMicroglia-Müller glia cell interactions control neurotrophic factor production during light-induced retinal degeneration.
Activation of microglia commonly occurs in response to a wide variety of pathological stimuli including trauma, axotomy, ischemia, and degeneration in the CNS. In the retina, prolonged or high-intensity exposure to visible light leads to photoreceptor cell apoptosis. In such a light-reared retina, we found that activated microglia invade the degenerating photoreceptor layer and alter expression...
